IMMUNOHISTOCHEMICAL STUDY ON GRANULAR CELL AMELOBLASTOMA

Abstract

The purpose of this study was to investigate the origin of the granular cells in granular
cell ameloblastoma and their pathogenesis by means of immunohistochemical reaction to
5primary antibodies. 7cases of granular cell ameloblastoma, 5 cases of follicular
ameloblastoma, 6 cases of tooth follicles were selected from the Department of Oral
Pathology, Seoul National University. They were routinely processed for
immunohistochemistry employing Avidin Biotin Complex methods. The primary
antibodies employed in this study were monoclonal cytokeratin, S-100, vimentin,
muramidase, and ¥á-1-antichymotrypsin. While the stellate reticulum cells in follicular
ameloblastoma showed strong positive reaction to cytokeratin antibody, the granular cells
showed moderate to slight positivity. Anti-S-100 protein antibody did not reacted with
the granular cells and other tumor cells. Some granular cells with small amount of
cytoplasm showed strong positive reaction to ¥á-1-antichymotrypsin antibody, other
granular cells with abundant eosiophilic cytoplasm showed moderate or no reaction.
Anti-mura-midase antibody and anti-vimentin anti-vimentin antibody did not reacted
with the granular cells.
We suggested that the granular cells were of epithelial origin and their cytoplasm had
reduced ¥á-1-antichymotrypsin positive granules in comparison with stellate reticulum
cells of follicular ameloblastoma.
